Abstract/Artist's Statement

In this project, a variety of organometallic compounds with Pt(II), Ru(II), or Ir (III) centers were synthesized in order to investigate their interactions with biomolecules with the goal of developing and investigating anticancer agents. Fluorinated ligands were targeted because they have shown promising results in their anticancer ability. Microwave synthetic techniques were employed to synthesize the complexes and they were purified by column chromatography. Two new fluorinated ruthenium (II) compounds were made, ND5400 and RU3FL and one new fluorinated platinum (II) complex, Pt3FL. The complexes were characterized by NMR and IR spectroscopies. Using the CRISPR-Cas9 system, the compounds were evaluated on their effects on enzyme activity and how they inhibit CRISPR. In addition, fluorescent studies were utilized to study if the compounds bind to DNA through groove binding mechanisms.
